Heavy lift launch vehicles are a supposition for the establishment of a Lunar Base and realizing Human Expeditions to Mars. Realistic proposals are available, however, detailed designs and optimization of heavy lift launch vehicles representing the current state-of -the-art are yet to be performed. Knowing the cost-effectiveness of individual system design parameters will greatly help to come up with a realistic design and viable program development proposals. The case study presented deals with a Lunar Space Transportation System comprised of a 3-stage Heavy Lift Launch Vehicle and a Space Operations Center in Lunar Orbit. The TRASIM computer code employed allows to analyse the influence of over 150 system parameters with 250 individual assumptions. Some of the more important variables with respect to system cost-effectiveness have been analysed with respect to development-, production- and operation cost, as well as specific transportation cost of passengers and cargo. Among those discussed in this paper Are: Annual launch rate, duration of program life cycle, subsystem reusability, spare rate.
