In this thesis, I attempt to identify and study the structures in the ISM in the constellations of Puppis and Vela (extending from l = 240 to 275 degrees and b = +/-20 degrees) within about 2 kpc. The early-type stars in the region were studied through their proper motions and a relation has been proposed to establish the connection between the early-type stars and the structures in the ISM. Three principal large-scale structures in the ISM were studied: (1) The Vela Molecular Ridge (VMR), a giant molecular cloud located between 1 to 2 kpc (2) the newly-discovered IRAS Vela Shell, a ring-like structure discovered (during the course of this thesis) on the IRAS ISSA maps, with a radius of 7.5 degrees centred around (l,b) = (263,-7). The Vela OB2 association stars lie within this shell -- in fact the shell envelopes these stars and are physically associated with the shell. The distance to this shell is 450pc. The cometary globules and dark clouds in the Puppis-Vela region are part of this shell and (3) the Gum Nebula a 36-degree diameter Hα structure. The OB associations most relevant for the interpretation of these three ISM structures were studied: (1) Vela OB1 at 1.5 kpc (2) Vela R2 at 800pc (3) Vela OB2 at 450 pc and (4) the O-type supergiant Zeta Puppis. Both proper motions and photometric data results showed Vela OB2 to be a relatively aged association (20 million years) on the verge of disintegration. The energy output of Vela OB2 was compared to the observed kinetic energy of the IRAS Vela Shell and is consistent with the IRAS Vela Shell being created by the Vela OB2 stars through stellar winds and supernovae. Zeta Puppis, a runaway OB star has a past projected path in the sky that strongly suggests that it originated in the VMR, close to the Vela R2 association. The structure and kinematics of the Gum Nebula are consistent with a shell structure located at the edge of a molecular cloud namely, the VMR. Therefore, the IRAs Vela Shell and the Gum Nebula are two separate structures which are seen overlapping in projection.
