The relative intensities of the emission lines H alpha, lambda 6584 of (NII), lambda-lambda 6717,6731 of (S2) for the nucleus, and 10 H2 regions of M101 are given. From the (S2) lines ratio, it was found that Ne=3000/cu cm in the nucleus, while Ne is less than 1000/cu cm in the H2 regions. The presence of a peculiar structure in the nucleus is revealed. Physical conditions and N/S abundance ratio in the H2 regions are very close to those obtained for M33. An upper limit for the surface brightness of the interarm area of M101 was established.
