A set of strict theoretical formulae for calculation of differential atmospheric refraction have been derived in this paper. Taking the LAMOST (Large sky Area Multi-Object fiber Spectroscopic Telescope) as an example, the effect of differential refraction on large field and long exposure observations is computed with these formulae. By comparison with results of other authors, the effect of the differential refraction calculated with the present method is more complete and independent of the type of telescope and guiding method. And then the effect of atmospheric dispersion on fiber spectroscopy is discussed and a method of reducing the effect is suggested.
