Arctic and Antarctic ancient permafrost sediments should be evaluated as Earth extremal environments that are very useful for improvement of methodologies for astrobiological search for life. The overwhelming majority of microorganisms revealed themselves as non-culturable and could be detected only by direct visual microscopic analysis of native samples. The up-to-date high resolution microscopy techniques offer new advantages for in-situ microbial investigations at the micron and submicron levels. Visualization of cells or cell populations directly in native samples allows a thorough analysis of the distribution of biogenic and abiogenic components in heterophasic media. Direct in situ revealing of microbial cells in native permafrost samples and cytomorphological analysis were carried out using epifluorescence microscopy, scanning electron microscopy (SEM) and atomic force microscopy (AFM) techniques, for fine cells structure investigations transmission electron microscopy (TEM) was applied. The application of environmental scanning electron microscope (ESEM) for studying intact permafrost samples has enabled us to reveal changes in the microstructure of frozen deposits upon their thaw. It was shown that atomic force microscopy techniques could provide differentiation in morphology of bacteria in depends of their physiological state. AFM approach gives also some possibilities for differentiation of biotic and abiotic objects in heterogeneous environment.
