The spectrophotometric data already available on giant H II regions in M101 are used to resolve the ionization structure of these objects. A rough approach to determine total helium and oxygen abundances by using the information on the spatial variation of ionic abundances yields Yp = .24 and delta Y/delta Z = 2.3. The radial variations of He(+) are found to be inconsistent with the behavior expected from the oxygen ionization structure. Ionization corrections for He obtained in the usual way may therefore be in error. The often incomplete coverage of these giant H II regions and related objects by small aperture observations possibly leads to systematic effects in the determination of chemical abundances.
